Settings > about phone > diagnostics > test > test the microphone, does it work? 

also boot on safe mode and see if it works while on safe mode.

I would do as @uliwooly suggests and check to see if this is a hardware issue using Xperia Diagnostics. If it fails the test have a chat with your contact your Local support team as it will need to be serviced.
